REMOVING PHENOLIC COMPOUNDS

LEACHING OR DISPERSAL

  • pretreatment : soak in running water

  • left in sterile water 2-3 hours after isolation

  • transfer to new medium (exudate becomes trapped by agar and becomes concentrated) [5-6 transfers at 2-3 day intervals until phenolic oxidation ceases]

  • Initial culture on liquid medium

ADSORPTION ONTO ACTIVATED CHARCOAL

  • absorbs inhibitory compounds (1-5g/l) from the medium, but also other ingredients (mainly organic molecules)

ABSORPTION BY POLYVINYLPYROLIDONE (PVP)

  • proteins, amides and polyamides can restore enzyme activity if prevented by polyphenols or tannins (caffeine = amide: PVP = polyamide often used with mercaptoethanol)

    PVP does not always work
