Chimeric shoots can be regenerated in vitro from callus that is composed of cells of different genotypes. Such callus can originate through induced mutation or by mixing callus of two (or more) genetically different plants. The induced shoots must have a multicellular origin.

grafting in vitro

In vivo as well as in vitro, cells from both rootstock and scion can contribute to the generation of an adventious shoot at the graft union. After a splice or a saddle graft has healed, all but a thin layer of the scion is removed. All adventitious shoots that are not arising from the graft union are removed.
