The next strides forward in the culture of plant tissue came with the advent of successful culture of wound tissue or callus from the excised cambial tissue of Salix by GAUTHERET in 1934. He used nutrient media solidified with agar but found that after 6 months the activity of the cultures ceased. In 1929 WENT discovered the growth regulator indoleacetic acid IAA which was shown by SNOW in 1935 to stimulate cambial activity. By including IAA and the vitamins B in his media GAUTHERET extended the culture period of the Salix callus to 18 months and was able to subculture. However after 18 months once again growth ceased.
