During the 1960s Cocking developed the enzymatic isolation and culture of protoplasts (Cocking, 1960). The method involves removing the cell wall with purified preparations of cellulase and pectinase, while regulating protoplast expansion with an external osmoticum. The cultured Protoplasts regenerate new cell walls, form cell colonies, and ultimately form plantlets. |
