Microspores


Pollen is removed from the anther, either by mechanical means or by natural dehiscence of the anther, and the isolated pollen is cultured on a liquid medium

Advantage of microspore culture
over anther culture:

  • In anther culture development of the microspore
    often does not progress past the first division due
    to inhibitory substances in the anther
  • Rapid proliferation of anther wall tissue can occur,
    resulting in non homozygous diploids
  • Starting with isolated microspores will always yield
    homozygous plants, even if they are diploids
    (case of different cereals) or even triploids (Petunia)
  • Microspore culture gives a homogenous population
    of plants resulting from developing microspores or
    pollen grains, while removing anther wall tissue
